目的 本文采用四氯化碳(CCl4)诱导的急性化学性肝损伤动物模型,评价3种植物功能盐(竹盐、松盐和梅盐)对实验小鼠急性化学性肝损伤的防护作用。方法 将45只5周龄的C57BL/6健康雄性小鼠适应性喂养7 d后随机分成9组, 末次给药 1 h 后,模型组和试验组小鼠均腹腔注射0.1 %的CCl4橄榄油溶液,剂量为10 mL/kg.bw,正常组给予等体积的橄榄油作为对照。禁食过夜,16 h后取血液和肝脏样本,测定血清中天冬氨酸转氨酶(AST)的和谷氨酸转氨酶(ALT)的活性; 测定肝组织匀浆中丙二醛(MDA)含量、以及谷胱甘肽(GSH)、总超氧化物歧化酶(T-SOD)和谷胱甘肽过氧化物酶(GSH-Px)的活力;同时,观察实验小鼠肝组织的病理损伤程度。结果 竹盐、松盐和梅盐均可不同程度地降低血清转氨酶(AST和ALT)的水平,降低肝脏的MDA含量、提高GSH水平及T-SOD活力,并减轻肝脏的病理性损伤。结论 表明以竹盐为代表的3种植物功能盐对CCl4诱导的小鼠急性化学性肝损伤均有一定的保护作用。
Abstract:
Objective This paper deals with the protective effects of three plant functional salts (bamboo salt, pine salt, and plum salt) on acute chemical liver injury in mice induced by carbon tetrachloride (CCl4). Methods Forty-five healthy C57BL/6 male mice were randomly divided into 9 groups after 7 days of adaptive feeding. One hour after the last administration, the mice in the model group and experimental group were intraperitoneally injected with 0.1% CCl4 olive oil solution at a dose of 10 ml/kg.BW, and the normal control with same volume of olive oil. All mice were fasted overnight, and blood and liver samples were taken after 16 h. The activity of serum glutamate transaminase(ALT) and aspartate aminotransferase(AST), the contents of malondialdehyde(MDA) and glutathione(GSH) in liver homogenate, and the activity of total superoxide dismutase(T-SOD) and GSH-px in liver homogenate were measured. At the same time, the pathological damage degree of liver tissues in mice were examined.Results Bamboo salt, pine salt and plum salts could reduce the activities of serum transaminase (AST and ALT), reduce the contents of MDA in liver, increase of GSH level and SOD activity, and alleviate the pathological damage of liver. Conclusion Three kinds ofplant functional salts all show certain functions on chemical liver injury in mice induced by CCl4.
